چکیده

Formiminoglutamic acid is a product derived from the metabolism of histidine by mammalian liver and several microorganisms (l-3). The metabolism of FGA’ is dependent upon the presence of tetrahydrofolic acid (4, 5), and consequently this formimino compound is excreted in the urine of human subjects or laboratory rats which have a deficiency of folic acid (6-8). Daft (6) has reported that suitable amino acid mixtures will partially replace the need for folic acid in the growth of the laboratory rat. Therefore, a study was undertaken to determine the influence of amino acids on the excretion of FGA in the urine of the rat. It was found that dietary methionine significantly reduces the excretion of FGA. We shall describe these observations and the implications derived therefrom.

Folate deficiency and formiminoglutamic acid excretion during chronic diethylnitrosamine administration to rats.

Elevated levels of the histidine catabolito formiminoglutamic acid were excreted into the urine of rats that were given both 0.01% diethylnitrosamine in their drinking water for 1 to 5 weeks and an injection of a loading dose of histidine. Similar histidine loading of control rats that received no carcinogen did not produce an elevation in urinary formiminoglutamic acid excretion. The elevation...

BRIEF REPORT Lack of Effect of L-Methionine Ingestion on 14C02 Excretion from L.Histidine (Imidazole.2.14C) in Folic Acid and Vitamin B12 Deficient Humans

A DIET CONTAINING ONE TO TWO PER CENT L-METHIONINE normalizes increased urinary excretion of formiminoglutamic acid ( FIGLU ) and decreased 14CO2 excretion from L-histidine ( imidazole-2-14C) in folic acid and vitamin B12 deficient rats. 2 If such findings could also be found in man, and if they reflect biochemical effects of methionine on monocarbon fragment metabolism rather than competition ...

Metabolism of formiminoglutamic acid by vitamin B12 and folic acid-deficient rats fed excess methionine.

The amidine carbon atom (C-Z) of L-histidine is known to be an efficient metabolic precursor of single carbon units. Following the administration of n-histidine-2-Cr4, the radioactivity has been found in CO2 and urea, in carbons 2 and 8 of purines, in the hydroxymethyl group of serine, and in choline, and creatine (l-4). The major pathway of histidine degradation which involves deamination to u...

Urinary excretion of formiminoglutamic acid by human subjects after antifolic acid therapy.

In the accompanying paper a sensitive and specific enzymatic method is described for the measurement of formiminoglutamic acid (1). In this paper data are presented on the application of this method for the assay of formiminoglutamic acid in the urine of a variety of patients. Formiminoglutamic acid is an intermediate in histidine metabolism, and requires tetrahydrofolic acid for its further me...
